Title: Simple and robust separation of monosaccharides in complex mixtures by capillary electrophoresis and high performance liquid chromatography

Overview: Plant fibre contains a complex mixture of carbohydrates that is partially liberated by acid hydrolysis. Hydrophilic Interaction Liquid Chromatography (HILIC), ligand-exchange liquid chromatography or other forms of LC with ion-exchange columns are popular techniques for analysis of acid hydrolysed plant samples. Capillary electrophoresis (CE) can also be used without derivatization at pH 12.6 with no sample pre-treatment other than dilution. Direct detection in CE of mono- and disaccharides is unexpectedly possible at 270 nm by photo-oxidation. This detection was investigated by NMR, CE and Photo-irradiation.
